The overall goal of this project is to develop a personalized platform for osteochondral (OC) regeneration by converging 3D printing technologies, biomimetic hydrogels, and bioactivity to replicating the complexity, specificity and function of native OC unit for OCD regeneration. The highly porous subchondral bone compartment will be extrusion 3D printed using gelatin methacryloyl (GelMA), incorporating bioactive nanofillers. The chondral compartment will be fabricated from polycaprolactone (PCL) melt electrowritten (MEW) fibrous scaffolds reinforcing a hybrid GelMA-PHEMA hydrogel, with deep, intermediate and superficial anatomical-inspired fibrous architectures, surface grafted for enhanced lubrication. PRP extracts will locally bioactivate for regeneration by recruiting and specifically differentiating MSCs as potential autologous bioactivation for personalized products (TRL 2 to TRL 4).
